Local & Desktop BLAST Options
You can BLAST your sequences against local custom databases using the Local and Desktop BLAST options. Create a database of sequences in text or FASTA format on your computer and BLAST your query sequence from Primer Premier. The results are then used for primer design. This support is especially useful if you do not want to submit your data on the Internet or if you are working with unpublished information.
手动引物搜索
Primer Premier为用户提供对引物设计的完全控制。使用手动搜索选项，用户可以选择模板序列上的任何位置来设计感测或反义引物。Primer Premier会立即计算并显示新引物的性能。然后在主窗口的序列视图中显示由用户终选择的引物。
Primer Premier软件还可以针对模板DNA 的来源以相应的遗传密码规则转换DNA 和序列。软件共给出八种生物亚结构的不同遗传密码规则供用户选择，有纤毛虫大核（Ciliate Macronuclear）、无脊椎动物线粒体（Invertebrate Mitochondrion）、支原体（Mycoplasma）、植物线粒体（Plant Mitochondrion）、原生动物线粒体（Protozoan Mitochondrion）、一般标准（Standard）、脊椎动物线粒体（Vertebrate Mito-chondrion）和酵母线粒体（Yeast Mitochondrion）。
